香草乙酮是多种精神药物和新型抗疟药的合成原料,在医学上常被用作强心剂和利尿药。它目前公认是一种特异性烟酰胺腺嘌呤二核苷酸氧化酶抑制剂,能有效减少再灌注损伤中的自由基生成。
作用机制香草乙酮通过抑制NADPH氧化酶的活性,从而减少其介导的自由基生成、中性粒细胞的浸润来保护心肌。这种保护作用能够减轻心肌再灌注损伤,有效治疗因自由基过度生成导致的心肌细胞凋亡。
应用香草乙酮因其特有的气味应用于香料工业,并作为有机合成原料用于生产精神类药物伊潘立酮和多种新型抗疟药。此外,它还是一种常用的抗氧化剂,在医学上常用作强心剂和利尿药。
制备将无水乙醇25 mL、愈创木酚2.0 mL(18.0 mmol)、无水碳酸钾5.0 g(36.2 mmol)、碘化钠0.3 g ( 2. 0 mmol)和溴代正丙烷3.2 mL( 35.4 mmol)依次加入反应瓶中,搅拌下回流反应10 h。过滤减压蒸除乙醇和过量的溴代正丙烷,加入二氯甲烷40 mL及1 mol-L-1氢氧化钠溶液30 mL,分液后有机相用水(2 x 25 mL)洗涤,无水硫酸镁干燥后旋蒸脱溶得黄色液体12.86 g, 收率95.3%。
在反应瓶中加入二氯甲烷25 mL和11.5 g ( 9.0 mmol),搅拌下加入无水氯化锌2.5 g( 18.3 mmol),冰浴冷却,滴加混合溶液[二氯甲烷(2.0mL)+乙酸酐(1.7 mL,18.0 mmol)]。滴毕缓慢升至室温反应4 h。加入1 mol-L-1盐酸40 mL分液,水相用二氯甲烷( 2 x 30 mL)萃取,合并萃取液用水(2× 30 mL)洗涤后无水硫酸镁干燥旋蒸脱溶得黄色液体混合物1.84 g, 收率97.9%,纯度99.1%。
在反应瓶中依次加入二氯甲烷16 mL及异构体3-甲氧基4-正丙氧基苯乙酮和3-丙氧基4-甲氧基苯乙酮的混合物1.0 g( 4.8 mmol),搅拌至完全溶解(5 min)。然后加入无水三氯化铝1.4 g (10.5 mmol)于35 ℃反应4 h。倒入1 mol-L-1氯化铵溶液25 mL中,分液后水相用二氯甲烷( 2 x 30 mL)萃取,合并萃取液旋蒸脱溶后经硅胶柱层析(洗脱剂:V(石油醚):V(乙酸乙酯)=10 : 1)分离纯化得米白色固体香草乙酮。
白色结晶性粉末。
