金荞麦为蓼科植物金荞麦 Fagopyrum dibotrys 的干燥根茎,具有清热解毒、排脓祛瘀的功效。其临床用于治疗肺痈吐脓、肺热喘咳及乳蛾肿痛等症状。研究表明,表儿茶素和原儿茶酸是金荞麦抗菌、消炎、镇咳的主要成分。金荞麦乙醇提取物中表儿茶素的含量占65%~70%,成为其含量测定的重要指标。此外,表儿茶素还对心脑血管疾病有防治作用,并能预防肿瘤和具有抗氧化功能。
研究显示,表儿茶素能够抑制LPS诱导的RAW264.7细胞炎症反应,其抗炎机制包括抑制细胞炎症因子NO、TNF-α、IL-1和IL-6的生成,下调iNOS蛋白表达,调控激活MARK信号通路中p38、ERK1/2和JNK蛋白表达。
抗氧化活性研究对金荞麦中(-)-表儿茶素活性物质的抗氧化性能进行了研究。超氧阴离子自由基清除实验结果显示其抗氧化率达到83.08%;羟自由基清除实验表明,活性物质质量浓度为0.350 mg/mL时清除率为77.30%,且质量浓度对清除率影响显著(P<0.01)。还原力测定实验显示其还原力达到同质量浓度VC的48.02%,同样表现出显著差异(P<0.01);乙醇提取物中活性物质含量达1.40 mg/mL,占块根干物质质量的2.52%。实验证明金荞麦(-)-表儿茶素活性物质具有良好的抗氧化性能,可作为天然高效的抗氧化剂。
乙醇提取工艺通过单因素试验法,并经F检验和新复极差法(SSR法)比较,确定了乙醇浓度、提取温度、料液比及提取时间的大致范围。进一步采用四因素三水平的正交试验,得到优化后的提取工艺为:乙醇体积分数60%、提取温度20℃、料液比1∶18、提取时间30 min,金荞麦(-)表儿茶素类活性物质的平均提取率达2.5%。该工艺简单合理,适用于金荞麦(-)表儿茶素单体产业化的前期生产。
