FMOC-赖氨酸可以通过以下步骤合成:以赖氨酸和氯甲酸-9-芴基甲酯为起始物料,在适当条件下进行一步反应制得。
首先,将干燥洁净的圆底烧瓶中加入赖氨酸的二氯甲烷溶液,并冷却至-5~5℃。然后,滴加三乙胺35 mL,继续搅拌半小时。随后,向反应体系中逐步加入配置好的氯甲酸-9-芴基甲酯的二氯甲烷溶液(89.4 g/200 mL),同样控制温度在-5~5℃,并搅拌40分钟。
接着,滴加3N盐酸溶液(300 mL)来终止反应,并将体系温度保持在0~10℃。继续搅拌半小时后进行分层操作。有机相用水洗涤三次后,在30-35℃下减压蒸除二氯甲烷。
然后加入石油醚(500 mL),并充分搅拌冷却至0℃,FMOC-赖氨酸将开始从溶液中析出。在0℃继续搅拌40分钟后进行过滤,并用石油醚(100 mL)洗涤滤饼。最后,在30~35℃下真空干燥后得到白色固体的FMOC-赖氨酸。通常该方法的收率在60~80%之间。
