Fmoc-L-苯丙氨酸是一种氨基酸衍生物,可用作手性药物及临床药物的中间体。
其合成方法如下:在0°C条件下,在二氯甲烷(15 mL)溶液中加入HCTU(0.64 g,0.0015 mol,1等摩尔量)、胺反应物(0.51 g,0.0013 mol,1等摩尔量)和DIEA(0.66 mL,0.0039 mol,3等摩尔量),随后加入fmoc-L-缬氨酸(0.44 g,0.0013 mol,1等摩尔量)。将反应混合物加热至室温并搅拌12小时。用二氯甲烷(DCM)(30 mL)稀释反应混合物,并使用NaHCO₃水溶液(2 × 20 mL)洗涤。在无水的Na₂SO₄下干燥有机层,然后真空浓缩。采用闪式柱层析进行纯化。
二肽反应物(500 mg,0.8 mmol,1当量)加入甲醇(22 mL),并添加钯/碳(100 mg,10 mol%)。在氢气(球形气球)气氛下搅拌反应1小时。过滤硅藻土,并用甲醇多次洗涤以合并滤液,最后在真空中浓缩。
将含有游离胺的聚合物加入到CH₂Cl₂(1.5 mL)和DMF(0.5 mL)中的Fmoc-AA₁-OH或AA₂-AA₁-OH二肽(1.2等摩尔量)及HCTU(1.2等摩尔量)溶液中,再加入DIEA(3等摩尔量),并搅拌反应3小时。将反应混合物浓缩至1 mL,加入乙醚(10 mL)。离心悬浮液,滗出上清液以得到含有支撑的三肽沉淀。
将沉淀溶解于DMF(0.5 mL)中,并用乙醚(10 mL × 3次)进行沉淀。在真空中干燥沉淀,重复上述去保护/偶联步骤(单个氨基酸),从而获得游离胺附着在载体上的肽。
使用LiOH·H₂O(4等摩尔量)于THF中将肽从载体分离1小时。上清液中的肽以盐的形式存在,而载体反应物则作为沉淀回收。用稀盐酸酸化多肽的锂盐,将其水溶液冻干。
最后,向DMF(0.5 mL)的载体溶液(50 mg,0.067 mmol,1等摩尔量)和Fmoc-AAOH(1.2等摩尔量)中加入N,N-二异丙基碳二亚胺(31.2 μL,0.2 mmol,3等摩尔量)和DMAP(2.4 mg,0.02 mmol,0.3等摩尔量)。在室温下搅拌反应6小时。与乙醚共沉淀后离心,得到目标产物Fmoc-L-苯丙氨酸。
