本品为白色或微黄色粉末,具有类似蒜的臭气,并有吸湿性。在水中或生理盐水中易溶,在乙醇、乙醚或丙酮中不溶。其密度为1.1335 g/cm³(20℃),熔点为-5℃,沸点约为146 - 147℃,折射率为1.53(20℃)。
简介辅酶A(Coenzyme A)是体内乙酰化反应的重要辅酶,在糖、脂肪及蛋白质的代谢中起重要作用。特别是在促进脂肪代谢方面更为关键。它能够激活体内的物质代谢,增强氧化作用并提供能量。主要用于治疗白细胞减少症和原发性血小板减少性紫癜,并可辅助治疗脂肪肝、急慢性肝炎、冠状动脉硬化、慢性动脉炎、心肌梗死、慢性肾功能减退引起的肾病综合征及尿毒症、新生儿缺氧、糖尿病和酸中毒等病症。
药理作用辅酶A是从鲜酵母中提取的,为体内乙酰化反应的重要辅酶。它可以与乙酸盐结合形成乙酰辅酶A,进而进入氧化过程,在糖、蛋白质及脂肪代谢中有重要作用。三羧酸循环、乙酰胆碱合成、肝糖原储存、胆固醇水平降低和血浆脂质含量调节等生理过程均与辅酶A密切相关。然而,目前也有观点认为,辅酶A的主要成分泛酸在人体中并不缺乏,一般患者无需补充辅酶A,并且外源性辅酶A为大分子不易通过细胞膜进入细胞内,其疗效存疑。
来源辅酶A由鲜酵母提取而成,主要组成包括泛酸、腺嘌呤、核糖、半胱胺及磷酸。
化学性质本品具有类似蒜的臭气和吸湿性,在水中或生理盐水中易溶,在乙醇、乙醚或丙酮中不溶。兼有核苷酸和硫醇的特性,是一种强酸。
用途辅酶A用于治疗白细胞减少症、原发性血小板减少性紫癜及功能性低热;并可用于脂肪肝、急慢性肝炎、冠状动脉硬化、心肌梗死、肾病综合征、尿毒症、新生儿缺氧、糖尿病和酸中毒等病症。此外,它还用于放射性损害的保护以及延缓肌肉萎缩的发展。
生产方法辅酶A从鲜酵母中提取。生产工艺步骤如下:新鲜压榨酵母(破壁)清液→一次吸附于树脂→吸附物洗脱→一次洗脱液二次吸附活性炭→吸附物洗脱→二次洗脱液浓缩还原→还原液络合→络合物净化→净化液浓缩沉淀→沉淀物干燥得到辅酶A。
