4-甲氧基苯硫酚属于一种芳香硫酚。芳香硫酚是一种具有广泛应用价值的有机硫化合物,广泛应用于医药、农药和染料等行业,还可以用于合成凝血酶阻聚剂、杀菌剂、皮炎药物、除草剂、缓蚀剂和感光材料等。
用途苯硫酚可用于生产医药氯霉素的代用品甲砜霉素及农药克瘟散等,并且可以制备橡胶再生剂、石油添加剂等。对氯苯硫酚是重要的农药和医药中间体,可用来合成增塑剂、油类添加剂和润湿剂;而4, 4′-二巯基二苯硫醚则可用于合成感光材料,是一种优良的光学树脂单体。
制备磺酰氯制备:向1000 mL三口烧瓶中加入0.25 mol芳香化合物苯甲醚和700 mL 1,2-二氯乙烷。开动机械搅拌并缓慢滴加3倍理论物质的量的氯磺酸,滴加过程中水浴控制底物温度约50 ℃,滴加完毕后升温至80 ℃保温反应1小时。反应结束后减压蒸除溶剂,所得固体经正庚烷重结晶得芳香磺酰氯纯品。中间产物的平均收率为85%。
芳香硫酚制备:取上述制得的芳香磺酰氯0.1 mol加入500 mL三口烧瓶中并加入200 mL正庚烷,开动机械搅拌。水浴控制底物温度85 ℃下分批加入3倍磺酰氯基团物质的量的三苯基磷。加完后继续在85 ℃反应1小时,随后降温至50 ℃,向体系中滴加100 mL水并继续反应10分钟。反应完毕后体系分层取油相用质量分数10%氢氧化钠溶液萃取(3次),取有机层减压蒸除正庚烷得到副产物三苯基氧磷保存待用;取碱层用质量分数37%盐酸酸化至pH=1~3,然后用二氯甲烷萃取(3次),取有机层用无水硫酸钠干燥,减压蒸除二氯甲烷即得芳香硫酚粗品。

An efficient approach for the direct sulfanylation of 4-hydroxyquinolinones and 4-hydroxypyridones with aryl thiols in the presence of CuI/DMSO has been developed. The substrate scope is broad, allowing facile synthesis of a range of structurally diverse 3-sulfanyl-4-hydroxyquinolinones and 3-sulfanyl-4-hydroxypyridones in good efficiency.
