依达拉奉是一种新型的自由基清除剂,于2001年4月在日本首次上市。主要应用于治疗缺血性脑卒中。其以阴离子形式存在,通过提供一个电子给自由基来清除自由基。由于分子结构中含有亲脂基团,易于通过血脑屏障进入脑组织,对脑缺血引起的神经细胞损伤具有保护作用。
作用机制依达拉奉对神经细胞的保护作用主要体现在以下几个方面:清除氧自由基、抑制脂质过氧化反应、调控炎症因子和抑制细胞凋亡。进一步研究表明,除了用于治疗急性脑缺血外,依达拉奉还对其他涉及氧化应激的神经系统疾病具有治疗效果,并且还能对非神经系统的其他器官产生保护作用。目前,依达拉奉主要应用于治疗缺血性脑卒中。
适应症适用于改善由急性脑梗死引起的神经症状、日常生活活动能力和功能障碍。
药物相互作用与头孢唑啉、哌拉西林和头孢替安等药物合用时,可能导致肾功能衰竭加重;与坎利酸钾或抗癫痫药地西泮、苯妥英钠等混合可产生浑浊现象。
制备方法一种依达拉奉的合成方法包括以下步骤:
白色结晶或粉末状物质。熔点为127℃,沸点在35.3kPa的压力下为287℃。溶于水和微溶于醇及苯中,不溶于醚、石油醚及冷水。
用途主要用于合成吡唑酮类解热镇痛药物(如安乃近、安替比林、氨基比林);亦作为吡唑酮类酸性染料(如酸性媒介枣红BN、永固黄G)、彩色胶片染料、农药及其他精细化学品的中间体。
用途用于制备吡唑啉酮染料和药物等。
用途用于测定钴、铜、铁、镍、银以及维生素B12,也可用于染料合成。
用途用于合成安替匹林、氨基比林、安乃近等药物;制备酸性媒介枣红BN、永固黄G、皮革喷涂红G等染料。
用途主要用于生产医药品如安替比林、氨基比林、安门静的原料,也用于染料及彩色胶片染料、农药及其他有机合成工业中,并可用作检测维生素B12和还原性碳水化合物的环保试剂。此外,也可用于检测还原糖类物质。
生产方法通过苯肼与乙酰乙酸乙酯反应制备,将两者在50℃左右加热并逐步加入乙酰乙酸乙酯,反应4小时后稍冷放置过夜析出黄色结晶,过滤纯化即可得到成品。另一种方法是使用苯肼和丁酮酰胺反应,在相同温度下进行反应,随后进行过滤、水洗和干燥处理以获取成品。原料消耗定额为:苯胺(98%)560kg/t、液氨1100kg/t、盐酸(30%)1280kg/t、醋酸1060kg/t、硫酸(93%)840kg/t、硫磺850kg/t、亚硝酸铵(95%)440kg/t。
