Fmoc-L-亮氨酸是N-fmoc保护的L-亮氨酸形式,为白色至灰白色的结晶粉末状物质。其密度为1.207 g/cm³,沸点在559.8°C (760 mmHg) 下测定,熔点则在152-156°C(分解)。
应用Fmoc-L-亮氨酸又称为Nα-9-芴甲氧羰基-L-亮氨酸,是一种氨基酸衍生物。它可以通过L-亮氨酸与9-芴甲氧羰基氯反应制备得到。有文献报道指出,该化合物可用于制备9-芴甲氧羰基氯。
制备具体制备方法如下:将1.05克(0.008摩尔)的L-亮氨酸固体溶解于10%碳酸钠溶液中,并在20~30℃下搅拌,直至完全溶解。随后,在此温度范围内,缓慢滴加2~205毫升甲苯中的9-芴甲氧羰基氯(2.10克,0.008摩尔)溶液,持续时间为30~60分钟。滴加完成后,在相同条件下继续搅拌1~8小时,然后加入30~200毫升水进行稀释,并使用乙酸正丁酯(80毫升)萃取除去过量的9-芴甲氧羰基氯。
所得水相需用浓盐酸酸化至pH值为0.5~3.5,再利用乙酸正丁酯(80毫升)进行萃取。最后,从油相中用水洗涤以去除残留的盐酸,并通过浓缩除去乙酸正丁酯溶剂,从而析出白色结晶产物Nα-9-芴甲氧羰基-L-亮氨酸2.46克,得率为87.0%,熔点为151~152℃。
