巨大戟醇是一种源自大戟科大戟属植物(如大戟、甘遂或千金子)的活性二萜类化合物。它最初于1968年从Euphorbia ingens中分离出来,2012年被美国食品药品监督管理局(FDA)批准用于治疗日光性角化病,并成为首个针对该适应症的药物。此外,针对基底细胞癌的II期临床试验也已完成。
提取来源巨大戟醇主要来源于大戟科大戟属植物的大戟全草、甘遂或千金子(大戟科植物续随子Euphorbia lathyris的种子)[2]。
分析方法通过高效液相色谱-紫外检测器(HPLC-DAD)或HPLC-电子捕获二极管检测器(ELSD)测定巨大戟醇含量,并辅以质谱(Mass)和核磁(NMR)鉴定。
药理药效具有显著的抗癌、抗病毒活性。
用途巨大戟醇可用于作为合成巨大戟醇甲基丁烯酸酯的关键中间体。2012年1月25日,Picato(商品名)获FDA批准用于光化性角化病(AK)的局部治疗[5]。
此外,HPLC-ELSD法测定巨大戟醇含量可作为甘遂饮片的质量控制方法[3]。
