在500 mL圆底烧瓶中加入10 g葡萄糖、100 mL乙酸、32.5 mL乙酸酐和0.5 g对甲基苯磺酸。超声反应10分钟后,向反应液中加入12.5 mL乙酰溴和9.0 mL甲醇,并在功率为420 W(型号DST20500)的条件下继续超声振荡1小时。TLC检测显示反应完全后,用二氯甲烷稀释并水洗,再通过乙醇重结晶得到21.9 g溴代葡萄糖晶体,产率为96%。
随后,在上述所得溴代糖中加入44 g β-环糊精、220 mL水和11 g锌粉。在功率为480 W(型号DST20500)的条件下进行超声波振荡反应10分钟后,用乙酸乙酯稀释并过滤。滤液经三次乙酸乙酯萃取后合并有机相,并加入无水硫酸钠干燥。最后通过减压浓缩和再结晶(使用乙酸乙酯和石油醚),最终得到13.8 g纯化的乙酰化葡萄烯糖,产率为95%。
