对溴苯乙炔可用作医药合成中间体。卤代苯乙炔是一类重要的电子化工原料和有机合成中间体,在药物、农药、催化、液晶中间体、阻聚剂、电极材料、发光涂料、腐蚀抑制剂等方面都有着广泛的应用。
应用(4-溴苯基)乙炔属于芳基末端炔,是一种烃类有机物。它主要用于有机合成,并且是重要的有机合成中间体。
用途通过芳基末端炔得到的聚芳炔、聚芳基二炔和芳炔类共轭大分子不仅在药物、农药等方面有着重要应用,而且在新型有机功能材料及分子电子器件中也有着广泛的应用前景。这些材料包括有机发光二极管、液晶材料、非线性光学材料、气体分离膜、分子导线、分子开关、分子计算机等。
制备 第一步反应在氮气保护下,向Schlenk反应瓶中依次加入三乙胺25 mL、4-溴碘苯1.415 g (5.0 mmol)、2-甲基-3-丁炔基-2-醇420 mg (5.0 mmol) 以及双(三苯基膦)二氯化钯(Pd(PPb)2Cl2)70 mg (0.1 mmol) 和三苯基膦(PPb)105 mg (0.4 mmol),碘化亚铜(CuI)38 mg (0.2 mmol)。反应在35℃下进行,直至薄层色谱监测原料对溴碘苯斑点消失为终点。
完成反应后,减压蒸干溶剂,并用CH2Cl2溶解。依次用5%稀盐酸、饱和NaCl溶液洗涤,无水MgSO4干燥,过滤。滤液减压蒸干后,粗产物以柱色谱提纯(洗脱液:石油醚∶乙酸乙酯=10∶1),最终得到白色固体4-(4-溴苯基)-2-甲基-3-丁炔基-2-醇1.052 g (4.4 mmol),产率为90%。
第二步反应在氮气保护下,向Schlenk反应瓶中加入甲苯30 mL、4-(4-溴苯基)-2-甲基-3-丁炔基-2-醇478 mg (2.0 mmol) 以及NaOH480 mg (12 mmol),并用二乙二醇单乙基醚8.6 mL。反应在75℃下进行,直至薄层色谱监测原料斑点消失为终点。
冷却至室温后,依次用5%稀盐酸、饱和NaCl溶液洗涤,无水MgSO4干燥,过滤。滤液减压蒸干溶剂后,粗产物以柱色谱提纯(洗脱液:石油醚),最终得到白色固体319 mg (1.76 mmol),产率为80%。
NMR谱图证明产物为(4-溴苯基)乙炔。
