简介

4-羟基环己酮乙二醇缩醛又称为1,4-二氧杂螺[4.5]癸烷-8-醇,可用作有机合成中间体。该化合物可由1,4-二氧杂螺[4,5]癸烷-8-酮还原得到。

制备

将带有温度计的250ml三口瓶中加入1,4-二氧杂螺环[4.5]癸烷-8-酮(10g,64.03mmol,等当量)、甲醇(80ml),搅拌直至完全溶解。在冰水浴下分两次缓慢加入硼氢化钠(共1.21g,0.032mol,50%过量)。加毕,移除冰水浴并继续搅拌0.5小时。通过薄层层析(TLC)检测(展开剂:DCM:MeOH = 100:1,显色剂:2,4-二硝基苯肼),反应完全。随后加入40ml水,并在减压下蒸除甲醇。使用乙酸乙酯进行萃取(50+50+25+25ml),合并有机相并用无水硫酸镁干燥,抽滤后减压蒸馏得到10.02g无色液体,收率98.9%。
