合成方法目前长春布宁主要通过半合成与全合成两种方式制备。半合成长春布宁主要是从马铃果中提取的它波宁为起始原料进行合成;而全化学合成法则通过中间体的还原获得。全化学合成法更适合工业化生产。
全化学合成路线全化学合成方法如下:(±)-15a-乙基-2,3,6,11,15,15a-六氢-1H,5H-吲哚并[2,3-a]吡喃[3,2-i]喹嗪-14,14-(13H)-二羧酸二乙酯是长春布宁化学合成过程中的中间结构。经手性拆分后进行还原,得到具有光学活性的(+)-15a-乙基-2,3,6,11,15,15a-六氢-1H,5H-吲哚并[2,3-a]吡喃[3,2-i]喹嗪-14,14-(13H)-二羧酸二乙酯(化合物Ⅰ),再经后续Pd-C反应得到长春布宁产品的有效结构(化合物Ⅱ)。
