制备乙酰苯胺时,将0.3 mol 苯胺、40 mL 冰醋酸和0.4 g 锌粉加入圆底烧瓶中,装上分馏柱,并在柱顶安装温度计。蒸出的水和乙酸收集于烧杯中。缓慢加热15 min后,在约105℃下进行蒸馏。当温度开始下降时,停止加热直至达到终点。趁热将反应物倒入冰水中,析出固体,冷却并抽滤以获得粗品乙酰苯胺。用水重结晶后得到白色纯2-甲基乙酰苯胺。
接下来制备2-氯-8-甲基喹啉-3-甲醛:将0.05 mol 2-甲基乙酰苯胺和0.15 mol DMF加入四口烧瓶中,安装温度计、机械搅拌器和冷凝管。在冰浴条件下以低于10℃的温度下滴加0.35 mol 的POCl₃,并于75℃下搅拌反应12小时。随着颜色从浅黄色油状液体变为橙红色液体,表明反应已经完成。停止加热后,在搅拌下将反应物缓慢倒入400 mL 冰水中,静置后抽滤析出的固体。用水洗涤三次并在空气中干燥。最后用乙酸乙酯重结晶以获得浅黄色针状晶体2-氯-8-甲基喹啉-3-甲醛。通过质谱、元素分析、红外和核磁共振光谱等方法对该产物进行表征,确认其为浅黄色针状晶体,产率为90%,熔点150~152℃。
