合成方法在加压圆底烧瓶中,将吡啶(2.66 mL,0.033 mol)加入黄嘌呤(5.00 g,0.033 mmol),搅拌后逐滴加入氯氧化磷(6.00 mL,0.066 mol)。随后加热至180°C 保持5小时。将反应生成的黑色溶液缓慢倒入冰水中(100 mL),搅拌20分钟后,用乙醚∶乙酸乙酯(5 x 20 mL,1:1)萃取有机物,并使用盐水(20 mL)洗涤,再通过硫酸钠干燥并浓缩,最终得到目标产物2,6-二氯嘌呤。产率为2.05克(0.011 mol),产率33%。黄色固体。(Rf=0.15;3%甲醇:DCM)。核磁共振数据如下:1H NMR(500MHz,DMSO-d6)δ8.70(1H,s),3.29(1H)。13C NMR(125 MHz,DMSO-d6)δ156.63、151.32、148.53、148.03、129.05。
2,6-二氯嘌呤用作抗肿瘤和抗病毒药物的中间体。
