β-阿法菌素(β-Apo-13-carotenone,D'Orenone)是天然存在的β-阿法菌素,能拮抗RXRα的激活。
体外研究β-apo-13-Carotenone 是在大鼠肠黏膜匀浆中 β-胡萝卜素酶解产物中被识别出的一种物质。它能够拮抗9-顺式视黄酸(9-cis-retinoic acid)对RXRα的激活作用,并且其效果在浓度低至1nM时仍显著。分子建模研究表明,β-apo-13-Carotenone 在分子层面上与RXRα的拮抗机制类似。它通过竞争性结合9cRA来抑制RXRα的活性,亲和力为7–8 nM,与9cRA本身相似。β-apo-13-Carotenone 的拮抗作用与已知的拮抗剂UVI3003具有类似的效力,能够诱导RXRα转录沉默四聚体形成,但不抑制共激活因子对LBD的招募。此外,β-apo-13-Carotenone 的摄取和/或代谢不会导致这些β-胡萝卜素代谢物在细胞中的积累。研究还发现,它能促进3T3-L1脂肪细胞标记基因的表达。
