应用广泛的3-甲氧基-4-氟苯甲醛是有机合成的重要原料,也是多种抗肿瘤药物和选择性环氧化酶-2抑制剂的重要中间体。
制备该化合物时,在装有温度计和滴液漏斗的100 mL三颈瓶中加入六次甲基四胺2.5 g(17.84 mmol)及三氟乙酸9 mL。开启搅拌,加热升温至80~84℃,在35 min内滴加邻氟苯甲醚1.12 g(1 mL 8.92 mmol)与三氟乙酸的混合液。维持该温度下继续反应1小时后进行常压浓缩回收溶剂,并稍冷后加入45 mL水,搅拌10分钟后用无水碳酸钾调节pH至7,然后使用50 mL乙醚萃取3次合并醚层。再用水(50 mL×3)洗涤醚层并用无水Mg2SO4干燥12小时。蒸净乙醚后得到黄色固体12 g,收率为87.3%。
