bardoxolone甲基是一种新型合成三萜类抗氧化抗炎调节剂,能强烈诱导Nrf2并抑制NF-κB和Janus激酶/STAT信号通路。研究表明,bardoxolone甲基可以促进癌细胞分化、抑制增殖并诱导凋亡。
体内研究bardoxolone甲基治疗的猴子肾切片显示,尽管各组mRNA表达相似,但巨噬细胞蛋白表达降低。这一减少在猴肾脏中的表达通过密度分析得到证实,表明bardoxolone甲基给药显著降低了猴肾脏中巨噬细胞蛋白的表达。与对照组相比,bardoxolone甲基治疗并未影响肾小管蛋白cubilin的表达。猴子接受bardoxolone甲基治疗后,血清肌酐清除率在第28天显著不同,并且与接受溶剂治疗的动物之间存在差异。24小时尿液中尿白蛋白/肌酐比值(UACR)在接受bardoxolone甲基治疗的猴子中显著增加,而对照组则下降了53.3%。
在HFD喂养期间口服bardoxolone(HFD/BARD)的小鼠、仅接受高脂饮食(HFD)的小鼠或低脂饮食(LFD)21周的小鼠中进行研究。与LFD小鼠相比,HFD小鼠的F4/80阳性冠状结构数量显著增加(+95%,P<0.001),而bardoxolone治疗有效预防了这一现象(-50%,P<0.01)。同样,HFD小鼠中F4/80间质巨噬细胞的数量显著高于LFD小鼠(98%,P<0.001)以及与HFD/BARD处理的小鼠相比(32%,P<0.01)。
