萘啶酮酸又称为萘啶狄克酸或温吐曼龙,是一种白色至淡黄色的结晶性粉末,无臭、无味。其熔点为229~230℃,难溶于氯仿、醇、水和乙醚。它是抗菌剂的一种,主要用于治疗由革兰氏阴性细菌引起的尿道感染,是喹诺酮类的第一代代表药物。
作用机制萘啶酸通过抑制细菌的DNA促旋酶,干扰DNA的合成,从而抑制生长期细菌的繁殖。其抗菌谱较窄,对大多数革兰氏阴性杆菌有效,但对铜绿假单胞菌(Pseudomonas aeruginosa)无效;同时对革兰氏阳性细菌和厌氧菌均无作用。
生物活性Nalidixic acid (NSC-82174) 是一种合成的1,8-二氮杂萘类抗菌剂,具有有限的抗菌谱,主要通过抑制细菌DNA促旋酶的A亚基来发挥作用。

用途该药物是一种抗感染药,对大肠杆菌、变形杆菌属等革兰氏阴性杆菌有一定的抗菌作用。主要用于治疗由革兰氏阴性细菌引起的尿道感染。萘啶酮酸是喹诺酮类的第一代抗菌药代表,第二代以吡哌为代表,第三代喹诺酮类的代表药物则是氟哌酸。
用途(重复)该品为抗菌剂,能抑制细菌DNA和RNA的合成,用于治疗由革兰氏阴性细菌引起的尿道感染。萘啶酮酸是喹诺酮类的第一代抗菌药代表,第二代以吡哌为代表,第三代喹诺酮类的代表药物则是氟哌酸。
用途此外,该药物还是一种细菌DNA合成酶(DNA gyrase)和鸟类成骨髓细胞瘤病毒反转录酶(avian myeloblastoma virus reverse transcriptase)的抑制剂。
生产方法萘啶酮酸由2-甲基吡啶制得2-氨基-5-甲基吡啶,再与原甲酸乙酯及丙二酸二乙酯缩合成N-(2-甲基-5-氨基吡啶)甲叉丙二酸二乙酯。然后将其在260~270℃环合后再加入氢氧化钠稀溶液中水解,得到7-甲基-1,8-萘啶-4-羟基-3-羟酸。最后用溴乙烷进行N-烷基化并异构成最终的萘啶酮酸。
