20(S)-Protopanaxadiol (PPD, 20-Epiprotopanaxadiol) 是人参皂苷在肠道的主要代谢产物之一,也是人参中的活性成分。它能在耐多种药物的癌细胞中抑制P-glycoprotein。
体外研究20(S)-Protopanaxadiol (PPD) 引起细胞生长抑制,在Int-407细胞中的LC50为23 μg/mL,而在Caco-2细胞中为24 μg/ml。在Int-407细胞系中,PPD 导致处于sub-G1(凋亡细胞)状态的细胞占比增多,这一反应与人参皂苷糖基化差异以及疏水亲水平衡有关。此外,PPD 还显著减少了U251-MG和U87-MG细胞的生存能力。其毒性作用伴随着细胞黏连蛋白表达减少,包括N-cadherin和整合素β1,导致黏着斑激酶磷酸化减少,并且还减少了cyclin D1的表达,从而诱导细胞在G1期阻滞。
体内研究在高架十字迷宫试验中,在束缚应激前处理以20(S)-Protopanaxadiol (PPD) 和PPT(20(S)-Protopanaxatriol),可以延长开臂滞留时间和开臂次数。处理以PPD 能有效抑制束缚应激所诱导的皮质脂酮和IL-6的血清水平,表明其可能通过GABAA受体和羟色胺受体发挥抗焦虑作用。与PPT相比,PPD 的抗炎作用更强。
化学性质20(S)-原人参二醇为白色结晶粉末,可溶于甲醇、乙醇、DMSO等有机溶剂,来源于人参。
用途用于含量测定/鉴定/药理实验等。药理药效方面,人参总皂苷及单体化合物对促进肿瘤细胞凋亡,促使肿瘤细胞分化,提高肿瘤细胞对化疗药物的敏感性,抑制肿瘤新生血管形成,提高机体抗肿瘤免疫力及抑制肿瘤生长和转移等方面具有重要作用。(20S)-原人参二醇是二醇组皂苷的代谢苷元,作为细胞凋亡诱导剂。
