3,4-二甲氧基苯乙酸甲酯又名高藜芦酸甲酯,可用于合成β受体阻滞药贝凡洛尔(bevantolol)和抗心律失常药维拉帕米(verapamil),也是合成异喹啉类生物碱的关键中间体。
制备3,4-二甲氧基苯乙酸甲酯的过程如下:在250毫升三颈瓶中加入3,4-二甲氧基苯乙酮酸乙酯(17.9克,0.075摩尔)、乙二醇(60毫升)和85%水合肼(22.1克,0.375摩尔),缓慢升温至120℃反应2小时。冷却至室温后加入氢氧化钾(12.6克,0.225摩尔),加热至180℃,边升温边蒸出低沸物约2小时,再回流反应4小时。反应结束后冷却至室温,加6摩尔/升盐酸(约30毫升)调pH值至2~3。加入水(150毫升),用乙酸乙酯萃取(50毫升×4次),合并有机相。用水洗涤两次(每次50毫升),经无水硫酸钠干燥后抽滤,滤液减压浓缩至干,剩余物用乙酸乙酯-石油醚(1∶1)重结晶,得到白色固体3,4-二甲氧基苯乙酸(10.8克,73.4%)。
将3,4-二甲氧基苯乙酸与甲醇进行酯化缩合即可制得目标产物3,4-二甲氧基苯乙酸甲酯。
此外,Methyl homoveratrate是RWJ-26240的体内代谢物,在血浆、尿液和粪便中均能检测到。McN5691(RWJ-26240)是一种电压依赖性钙通道阻滞剂。
