应用
N-苄氧羰基-D-缬氨酸是一种氨基酸衍生物,可通过D-缬氨酸与氯甲酸苄酯反应制备。常用的碱是碳酸氢钠和氢氧化钠。
制备
在0℃下,将10 mmol D-缬氨酸溶解于50 ml碳酸氢钠(NaHCO₃)饱和溶液中,得溶解液。边搅拌边以每10秒滴加一滴的速度加入11 mmol的氯甲酸苄酯进行反应。滴完后,置于30℃下继续搅拌反应15小时,并做薄层检测。待溶液完全反应后,用50 ml/次乙醚洗涤三次,弃去乙醚,得到水相。
将水相调pH至3(出现乳白色固体),然后使用50 ml/次的乙酸乙酯萃取三次,合并所得液体。接下来依次用水、饱和食盐水各洗一次,再用无水硫酸镁干燥7小时后抽滤。滤液减压浓缩得到淡黄色油状物。
以体积比为1∶3的乙酸乙酯和石油醚配制的混合溶剂作为流动相进行层析柱纯化,最终得到N-苄氧羰基-D-缬氨酸,产率为68.7%,呈无色透明油状。
