埃博霉素(Epothilones)是黏细菌纤维堆囊菌产生的大环内酯类次级代谢产物,是一种新型的抗肿瘤药物。其作用机制与紫杉醇相同,通过促微管聚合作用从而抑制肿瘤细胞的增殖。相较于紫杉醇,埃博霉素具有毒性更小、水溶性更好、对紫杉醇耐药性细胞作用更强、抗肿瘤谱更广等优点,是最具潜力的抗肿瘤新药之一,目前在结肠癌、乳腺癌和卵巢癌治疗中取得了显著效果。
埃博霉素B结构式将保藏在固体斜面培养基的菌种接入放有已灭菌滤纸片的 CNST 平板上,倒置于恒温培养箱中,在 30℃条件下培养 5-7 天后,转接于 M26 培养基中。装液量为 50 mL/250 mL 三角瓶,在 30℃、170 r/min 的条件下摇床培养 72 h 后得到作为发酵培养的种子液。
发酵培养:以 5%(v/v)的比例将种子液加入到初始发酵培养基中,进行发酵培养。在 30℃下摇床培养,监测并调整 pH 和营养成分。
研究 体内研究为了评估埃博霉素 D (Epothilone D, EpoD) 对 PS19 小鼠 MT 和轴突功能的影响,在 3 个月大的雄性 PS19 小鼠中每周腹腔注射一次溶剂或 EpoD(1 mg/kg 或 3 mg/kg),共持续 3 个月。此外,同窝的野生型 (WT) 小鼠也接受了相同剂量的 EpoD 或溶剂注射。结果表明,EpoD 的低剂量在这些研究中表现出良好的耐受性。所有接受药物治疗的小鼠体重增长与溶剂处理组无显著差异,器官相对重量相似。使用标准转棒测试评估的运动性能亦无明显差异,且白细胞计数和中性粒细胞含量之间也没有显著差异。
体内研究EpoD(1 mg/kg 或 3 mg/kg)每周腹腔注射一次共 3 个月,在 3 个月大的 PS19 小鼠和野生型 (WT) 小鼠中进行。结果表明,接受 EpoD 治疗的小鼠没有表现出药物不耐受的迹象,并且体重增长与溶剂处理组无显著差异。器官相对重量相似,运动性能评估亦未显示有明显差异。尽管存在轻微的群体间差异,但治疗组之间白细胞计数和中性粒细胞含量均无显著差异。因此,这些研究使用的 EpoD 低剂量似乎被良好耐受。
结论EpoD 在高剂量下可引起一些副作用如中性粒细胞减少症等,但在较低剂量下则表现出良好的安全性和有效性,在未来的临床试验中值得进一步探索和验证。
