甲氧苄啶是一种亲脂弱碱性的乙胺嘧啶类抑菌剂,又称磺胺增效剂、甲氧苄氨嘧啶、抗菌增效剂、甲氧苄胺嘧啶、甲氧苄嘧啶、三甲氧苄胺嘧啶或三甲氧苄啶。它在常温下为白色或类白色的结晶性粉末,无臭且味苦。该药物微溶于乙醇和丙酮,几乎不溶于水,在氯仿中略溶,并且极易溶解于冰醋酸中。
抗菌谱与磺胺药相似但作用较强,对大肠杆菌、奇异变形杆菌、肺炎杆菌、腐生葡萄球菌以及多种革兰阳性或阴性细菌有效。尽管对绿脓杆菌感染无效,其最低抑菌浓度通常低于10 mg/L。单独使用时易导致细菌产生耐药性,因此一般不单独使用。
甲氧苄啶主要与磺胺类药物联合使用,形成复方制剂,用于治疗尿路感染、肠道感染、呼吸道感染、菌痢、肠炎、伤寒、脑膜炎、中耳炎、流行性脑脊髓膜炎(流脑)、败血症及软组织感染等疾病。对某些细菌引起的感染如伤寒和副伤寒,其疗效不低于氨苄西林;也可与长效磺胺类药物联合用于防治耐药恶性疟疾。
抗菌作用原理甲氧苄啶的抗菌作用机制是通过抑制细菌二氢叶酸还原酶活性来干扰其叶酸代谢。它与哺乳动物的二氢叶酸还原酶相比,结合更为紧密(约5~6万倍),从而阻止了细菌核酸和蛋白质合成。
与磺胺药物联用时,会双重阻断细菌的叶酸合成途径,产生协同作用,增强磺胺药的抗菌活性,并使抑菌效果转化为杀菌效果。此外,甲氧苄啶还可增强多种抗生素(如四环素、庆大霉素等)的抗菌活性。
甲氧苄啶溶液配制方法 配置选择性培养基三甲氧基苯甲醛经缩合、环合并最终生成甲氧苄啶。原料为三甲氧基苯甲醛,先与甲氧基丙腈缩合形成3'4'5'-三甲氧基-2氰基-3甲氧基丙烯,再在甲醇/甲醇钠体系中与硝酸胍环合成。
用途 抗菌增效剂主要用于联合磺胺药物治疗细菌感染。也可作为兽药使用,用于治疗禽类大肠杆菌引起的败血症、鸡白痢、禽伤寒、霍乱以及呼吸系统继发性细菌感染等疾病,并可用于球虫病的治疗。
研究用途甲氧苄啶是一种具有原核生物酶选择性的二氢叶酸还原酶抑制剂,适用于特定剂型的研究中。它通过抑制四氢叶酸合成,针对原核生物中的特定二氢叶酸还原酶(DHFR)发挥作用。
