6-氯靛红是一种靛红衍生物。靛红(iSAtin, ISA)又名吲哚满二酮或2,3-吲哚醌,是存在于哺乳动物组织和体液中的内源性活性物质。
制备在40℃下,分批将无水硫酸钠(35g,246mmol)加入到含水合氯醛(5g,30.5mmol)的90ml水中。随后滴入含有3-氯苯胺(2.57g,27.6mmol)和36%浓盐酸(3ml,36mmol)的20ml水溶液,再滴入盐酸羟胺(6.1g,88.4mmol)的27.5ml水溶液。滴加完毕后升温至85℃反应2小时,冷却到50℃,抽滤、用冷水洗涤滤饼并烘干,得到N-(3-氯苯基)-2-肟基乙酰胺。
将98%浓硫酸(46ml)加热至60℃,在搅拌下分批加入N-(3-氯苯基)-2-肟基乙酰胺(1.8g,11mmol),于0.5小时内加完。加毕升温至80℃反应15分钟,冷却至室温后倒入250g碎冰中,在室温下搅拌0.5小时,抽滤并用冰水洗涤,得到橘黄色固体(1.2g,收率75%)。1H NMR(400MHz, DMSO-d6):6.72(m, 1H),6.85(m, 1H),7.59(m, 1H),11.17(s, br, 1H)。
