卡洛芬是一种白色结晶性粉末,不溶于水但易溶于乙酸等有机溶剂。
应用卡洛芬是一种非甾体类抗炎药,主要用于缓解老年犬关节炎症状。尽管其在兽医领域应用广泛,现已不再用于人类药物使用。
不良反应卡洛芬通常耐受良好,大多数不良反应较为轻微,如胃肠疼痛和恶心,这些症状类似于其他非甾体类抗炎药所引起的副作用。
生物活性Carprofen 可抑制犬科COX2,IC50为30 nM。

Carprofen(S和R立体异构体)能够抑制犬科COX2,IC50值为0.102微摩尔。其中,主要是S对映体比R对映体表现出更高的抑制作用,IC50分别为0.0371微摩尔和5.97微摩尔,约相差200倍。Carprofen与人血清白蛋白(HSA)的结合常数通过荧光和平衡透析法测定为K1 = 5.1 μM (荧光) 和3.7 μM (ED), K2 = 37 μM (荧光) 和13 μM (ED)。结合主要发生在位点II,苯二氮卓类受体位点;而Carprofen的低亲和力位点是位点I,华法林位点。其羧基在与HSA高亲和力结合中起到重要作用。
体内研究手术前使用Carprofen(4 mg/kg)比其他药物具有较低的疼痛评分,在拔管2小时后效果显著。Carprofen处理犬类后,导致最大血药浓度及曲线下面积均显著升高,并且第一时刻的曲线下面积也更高。与扑热息痛相比,Carprofen在止痛方面略胜一筹,产生较少镇静作用。此外,Carprofen处理犬类18小时都能提供良好的镇痛效果并且没有不良副作用,还能大大增加瘸鸟的速度。
化学性质卡洛芬为白色或类白色结晶性粉末,熔点范围为197-198℃。
用途用于消炎、镇痛。 适用于治疗类风湿性关节炎、骨关节炎、急性痛风及关节外风湿病等引起的疼痛。
