S- (+)-Rolipram 抑制人体单核细胞中环AMP特异性的PDE4,IC50为0.75 μM。它作用于中枢神经系统,具有消炎和抗抑郁的活性,但其效果比R-对映体略弱。
体外研究S-(+)-Rolipram 通过抑制PDE4,减少了人单核细胞中由LPS诱导的TNFα表达(IC50为2 μM)。在浓度为1 μM时,它显著抗卵清蛋白(OA)诱导的气管软骨环收缩。此外,在稳定表达重组全长人PDE-4a的CHO-K1细胞系中,S-(+)-Rolipram 抑制了PDE4活性(IC50为450 nM)。使用Rolipram处理的人胶质瘤细胞系A-172表现出p21(Cip1)和p27(Kip1)表达的增加,并降低了cdk2活性,导致细胞周期停滞在G1期。最终,这些细胞经历分化并发生凋亡。
体内研究S-(+)-Rolipram 处理麻醉的对OA敏感的豚鼠,可降低OA诱导的支气管收缩(ID50为0.25 mg/kg),但对静脉注射组胺和白三烯D4诱导的支气管收缩无明显效果。高剂量(3-10 mg/kg)处理后,S-(+)-Rolipram 降低了组胺而非白三烯D4诱导的支气管收缩。在有意识的对OA敏感的豚鼠中,使用S-(+)-Rolipram 进行胃部预处理可以降低OA诱导的具体电导下降和肺嗜酸性粒细胞涌入,并且这种作用表现出剂量依赖性。
