十一烯酸乙酰乳香酯(Acetyl-11-keto-β-boswellic acid,AKBA) 能显著抑制 5-脂氧合酶(5-LO)的活性,从而发挥强大的抗炎和抗氧化作用。此外,AKBA 对肿瘤、溃疡和免疫调节等疾病表现出良好的治疗前景。它可通过抑制 TNF-α 诱导的 NF-κB 活化,进而抑制基质金属蛋白酶的活化,起到血管保护作用;同类化合物甘草酸和积雪草酸被报道具有良好的抗氧化作用,通过调节体内不同细胞因子表达有效抑制和逆转血管重构。AKBA 是西黄丸药制剂中的重要活性成分。
采用薄层色谱法和高效液相色谱法建立西黄丸中乳香类成分——十一烯酸乙酰乳香酯的方法。通过高效液相色谱法对其含量进行测定,AKBA 含量范围为 0.27% 至 1.05%。薄层色谱法和高效液相色谱法可定性定量检验西黄丸中的十一烯酸乙酰乳香酯,并作为西黄丸现行法定检验标准中乳香显微鉴别的有益补充。
药理应用AKBA(Acetyl-11-keto-β-boswellic Acid) 是乳香提取物的主要药效成分之一。在一定质量浓度下,AKBA 可诱导 HL-60 细胞凋亡,并且其作用呈剂量和时间依赖性。KBA 作为乳香的单体成分,诱导白血病细胞凋亡作用强,在急性髓系白血病治疗领域具有良好的临床应用前景。
参考文献AKBA(Acetyl-11-keto-β-boswellic Acid)具有多种生物活性。在体外实验中,它能显著抑制人类结肠腺癌的生长,并导致细胞周期停滞于 G1 期及诱导凋亡;同时,在 3T3-L1 脂肪细胞中触发显著脂解作用,表现为细胞质中中性脂肪减少、培养基中游离脂肪酸增加,伴随脂解酶活性增强和脂滴稳定性调节蛋白 perilipin 表达下调。此外,AKBA 还降低了成熟脂肪细胞的 aP2、瘦素及葡萄糖-4受体表达。
体内研究在体内实验中,AKBA 能显著预防肠腺瘤形成且不具毒性;其预防小肠和结肠息肉的能力优于阿司匹林。组织病理学检查显示,AKBA 对较大息肉及其起源的结肠息肉具有更明显的效果。AKBA 通过口服给药能有效延缓 HT-29 肿瘤异种移植生长且无毒性;与阿司匹林相比其活性更强。AKBA 在小鼠中的口服应用显著抑制 SGC-7901 和 MKN-45 肿瘤的生长,且未见毒性。
用途用于含量测定、鉴定及药理实验等。
