PDE4选择性抑制剂Rolipram能以相似的方式抑制免疫纯化的PDE4B和PDE4D活性,IC50约为130 nM和240 nM。相比之下,它对PDE4A的抑制作用显著较低,IC50约为3 nM。Rolipram以剂量依赖性方式增加U937细胞中cAMP响应元件结合蛋白(CREB)的磷酸化,这表明存在高亲和力(IC50约1 nM)和低亲和力(IC50约120 nM)成分。此外,Rolipram以剂量依赖性且单调的方式抑制IFN-γ刺激下的p38 MAPK的磷酸化,其IC50约为290 nM。Rolipram是一种选择性的PDE4抑制剂,能够抑制所有PDE4同工型A、B、C和D。在J774细胞中以剂量依赖性方式抑制LPS诱导的TNF产生(IC50 25.9 nM),最大/亚最大抑制效应出现在2 μM药物浓度下。
体内研究在野生型小鼠腹腔巨噬细胞(PM)中,LPS诱导TNF mRNA和蛋白表达增加74%和63%,而Rolipram可以明显地抑制这种表达。与野生型相比,MKP-1(-/-)小鼠的LPS诱导的TNF产生增强,这与已发表的结果一致。有趣的是,在MKP-1(-/-)小鼠中,Rolipram对TNF mRNA和蛋白表达的抑制作用显著减弱,并未达到统计学意义。重复给予Rolipram(1.25 mg/kg,i.p.)可减少学习无助大鼠的逃避失败次数。
