应用5-氟吲哚是一种吲哚类化合物,可通过两步制备得到:首先从5-氟-2-硝基甲苯(化合物1b)出发,经过一系列反应步骤,最终用于有机合成和医药中间体的合成。它主要用于实验室研发过程及化工医药合成过程中。
制备具体制备方法如下:
将7.2克5-氟-2-硝基甲苯(化合物1b,0.046mol)和27.4克DMFDMA(0.232mol)加入到80毫升乙腈中。反应温度升至85℃~95℃后回流5小时。冷却反应液并旋干溶剂,用水和微量乙醇洗涤得到粗品产物2b(0.0443mol,收率96.2%)。
接下来,将所得中间体2b加入到147毫升含10wt%肼的甲醇溶液中(肼含量为0.45摩尔,即10当量)。将反应混合物升温至50℃,反应8小时后冷却至室温。旋干溶剂即可得到产物3b,即5-氟吲哚(399-52-0),其产量为5.7克(0.0424mol,收率95.8%)。
