合成化合物 9-2
将化合物 9-1(11.0g,44.8mmol)溶解在 DCM(200mL)中,在-78℃下缓慢滴加乙基二异硫氰酸酯 (Et2NSF3) 溶液(8.85mL,67.3mmol)。滴毕后,在该温度下反应2小时,随后在室温下继续反应19小时。当反应完全时,用氯化铵水溶液(100mL)淬灭反应,水层用DCM(100mL×3)萃取,合并有机相并用饱和食盐水洗涤,再使用无水 Na2SO4干燥后浓缩,通过柱层析分离纯化(洗脱剂:PE/EtOAc(v/v)=20/1),得到淡黄色液体7.75g。产率为70%。
合成化合物 9-3
将化合物 9-2(5.83g,23.6mmol)溶解在THF(30mL)中,在0℃下缓慢滴加氢氧化锂的水溶液(1.98g, 30mL)。滴毕后,在室温下反应2小时。当反应完全时,用稀盐酸(1M)调节反应液pH值至5,除去THF后,水层用稀盐酸(1M)调节pH值至2,然后用乙酸乙酯(EtOAc,80mL×3)萃取。合并有机相,使用饱和食盐水洗涤并经无水Na2SO4干燥后浓缩,最终得到白色固体5.27g。产率为96%。
