2-氯-7-甲氧基-3-喹啉甲醛的制备如下:将N-对甲苯基乙酰胺(0.05 mol)溶于9.6 mL二甲基甲酰胺(0.125 mol)中,在0℃下逐渐加入32 mL三氯氧磷(0.35 mol)。随后,将反应混合物置于配备有干燥管的回流冷凝器的圆底烧瓶中,并在75-80℃油浴上加热4-16小时。待冷却至室温后,再倒入100 mL冰水中。通过过滤收集形成的沉淀,并从乙酸乙酯中重结晶得到2-氯-7-甲氧基-3-喹啉甲醛,产率为75%;熔点为122-123℃(文献报道为123℃)。其红外光谱(FT-IR ATR)特征值如下:波数(cm⁻¹)3051(芳香族C-H),2873(醛C-H),1686(C=O),1576(C=N),1055(C-Cl)。核磁共振氢谱(1HNMR,400 MHz,氯仿-d)显示:δ 10.55(s, 1H),8.66(s, 1H),7.96(d, J=8.5 Hz, 1H),7.79-7.63(m, 2H),2.57(s, 3H)。质谱(MS-API)测定值为[M+H]+ 206.02,计算值为205.03。
应用2-氯-7-甲氧基-3-喹啉甲醛可用作医药合成中间体。
