Fmoc-L-异亮氨酸是一种Nα-9-芴甲氧羰基-氨基酸。9-芴甲氧羰基是肽合成中的一个重要氨基保护基,因为它是碱敏感的保护基且在脱去常用保护基条件下稳定,因此它在多肽合成中具有独特的作用。
应用9-芴甲氧羰基氯和氨基酸是合成Nα-9-芴甲氧羰基-氨基酸的重要原料。而Nα-9-芴甲氧羰基-氨基酸则是合成多肽药物的关键成分之一。
制备将1.05克(0.008摩尔)L-异亮氨酸固体溶解于10%碳酸钠溶液中,搅拌使固体充分溶解。在20~30℃下,滴加9-芴甲氧羰基氯(2.10克,0.008摩尔)溶于甲苯的溶液,滴加时间控制在30~60分钟。滴加结束后,在同一温度下搅拌1~8小时,然后加入30~200毫升水稀释,使用乙酸正丁酯(80毫升)进行萃取以去除过量的9-芴甲氧羰基氯。
所得水相用浓盐酸酸化至pH值为0.5~3.5,再用乙酸正丁酯(80毫升)进行萃取。油相用水洗涤除去盐酸后浓缩,并除去乙酸正丁酯溶剂。结晶析出后过滤并干燥,最终得到Fmoc-L-异亮氨酸2.48克,收率为87.9%,熔点为145~146℃。
