冬凌草提取物中的冬凌草甲素(又称冬凌草素、延命草宁),是从唇形科香茶菜属植物冬凌草的叶子中提取出的一种贝壳杉烯二萜类化合物。它具有清热解毒、消炎止痛、健胃活血及较好的抗肿瘤活性作用,现已发现其对肺癌、前列腺癌、乳腺癌等多种细胞株均有抑制作用。
用途:冬凌草甲素具有抗肿瘤、免疫抑制、抗病毒、抗早孕以及改变皮肤表皮的角化过程等作用。临床用于恶性肿瘤、银屑病、治疣、急慢性白血病以及血吸虫病引起的肝脾肿大等。
生物活性:Oridonin(Isodonol, Rubescenin, NSC-250682)是从Rabdosia rubescens中分离的对映贝壳杉烷二萜,是一种具有抗肿瘤、抗细菌和抗感染效果的传统中药。研究发现,Oridonin抑制AKT1 和 AKT2的激酶活性,对应的IC50值分别为8.4 μM 和 8.9 μM。

体外研究:Oridonin有效抑制各种癌细胞的增殖,包括来自前列腺癌(LNCaP, DU145, PC3),乳腺癌(MCF-7, MDA-MB231),非小细胞肺癌(NSCL)(NCI-H520, NCI-H460, NCI-H1299),急性早幼粒细胞白血病(NB4),以及胶质母细胞瘤(U118, U138)的细胞增殖,ED50s的范围为1.8至7.5 μ克/毫升。Oridonin的抗肿瘤活性可能是由于它对NF-κB和MAPKs信号通路的抑制作用。
体内研究:LD50: 小鼠35-40毫克/千克。
