5-氨基吲哚是一种吲哚类芳香胺。芳香胺类化合物是重要的有机合成中间体和原料,广泛用于农药、医药、染料、表面活性剂以及感光材料的合成。
制备将16.44毫克(0.04毫摩尔)4,4-二甲氧基-2,2-联吡啶银、11.22毫克(0.1毫摩尔)叔丁醇钾和1毫升1,4-二氧六环加入高压釜中,搅拌均匀后加入1毫摩尔5-硝基吲哚,在氢气压力为4.0兆帕下于80℃反应24小时。待反应结束后,将反应液用水与二氯甲烷萃取,收集有机相并经无水硫酸钠干燥、抽滤和旋转蒸发处理。最后通过柱层析分离得到5-氨基吲哚,产率为98%。其波谱数据如下:
1H NMR(400兆赫兹,CDCl3):δ 7.96(宽而烂,1H),7.20(d,J=8.4 Hz,1H),7.14(s,1H),6.95(s,1H),6.67(d,J=8.4 Hz,1H),6.38(s,1H),3.51(宽而烂,2H); 13C NMR(100兆赫兹,DMSO-d6):δ 141.0,129.8,128.5,124.7,111.8,111.4,103.2,99.6; HRMS m/z(ESI):理论值C8H9N2 [M+H]+=133.0760,实测值=133.0755。
