4-乙炔基苯甲醛是有机合成中间体和医药中间体,在实验室研发中的有机合成反应以及医药研发过程中都有应用。
制备化合物 4 的合成:将对溴苯甲醛(1.85g,10mmol)溶解在15mL无水THF中,加入三甲基硅基乙炔(1.47g,15mmol)、双三苯基磷二氯化钯(70mg,0.1mmol)、碘化亚铜(38mg,0.2mmol),以及5mL 三乙胺。在40℃反应2小时后旋干THF,以正己烷和二氯甲烷(体积比2:1)为洗脱剂过柱纯化,得到2.0g化合物4,白色固体。产率为99%。
化合物 5 的合成:将化合物4(727mg,3.6mmol)溶解在15mL无水THF中,加入2mL KOH水溶液(298mg,4mmol)。常温下反应2小时后旋干THF,反应液用乙酸乙酯萃取,并用饱和NaCl溶液洗涤三次。有机相经无水Na2SO4干燥、过滤和浓缩,以正己烷和二氯甲烷(体积比1:1)为洗脱剂过柱纯化,得到450mg化合物5,淡黄色固体。产率为97%。
