Skin contact: Immediately wash skin with copious amounts of water for at least 15 minutes while removing

化学性质

用途

2-氨基-3-氟苯腈主要用作有机合成和医药化学中的中间体,并广泛应用于基础化学研究及医药分子修饰中。其结构中的氨基和氰基作为易衍生化的官能团,在简单的反应条件下可以转化为叠氮基团、酰胺或羧酸衍生物,因此在合成制备含氟原子的1,2,3-三取代苯类化合物方面有着一定的应用。

合成方法

将干燥的500毫升圆底烧瓶中加入(Z)-7-氟-3-(异亚硝基)吲哚啉-2-酮(3.12 g,17.32 mmol),然后用二甲基甲酰胺(150 mL)配制成棕色/琥珀色溶液。将此反应溶液加热至180℃,维持稳定的回流并确保内部温度在152 °C,搅拌反应3小时后,在室温下继续搅拌过夜。反应结束后分别使用水(125 mL)、饱和碳酸氢钠水溶液(125 mL)和乙酸乙酯(250 mL)稀释反应混合物,摇匀并分离有机层与水层。其中水层用乙酸乙酯萃取两次,合并所有有机物后,用水反萃取一次进行干燥处理。使用无水硫酸镁干燥滤液后,在真空下浓缩残余物,并以二氯甲烷作为洗脱剂通过硅胶柱色谱法分离纯化得到目标产物2-氨基-3-氟苯腈。最终收率为78%(2.36 g,17.34 mmol)。
