4-羟基-7-苯氧基异喹啉-3-甲酸甲酯可用作医药合成中间体。首先由苯酚为原料制备中间体5-苯氧基异苯并呋喃-1(3H)-酮,再通过进一步反应生成2-(氯甲基)-4-苯氧基苯甲酸甲酯。最后,该化合物与N-对甲苯磺酰基甘氨酸甲酯进行反应得到4-羟基-7-苯氧基异喹啉-3-甲酸甲酯。
制备 第一步:5-苯氧基异苯并呋喃-1(3H)-酮将20.4g苯酚、50mL二甲基亚砜(DMF)、34.0g 5-溴异苯并呋喃-1(3H)-酮、3.2g乙酰丙酮、3.6g溴化亚铜和30.8g碳酸钾加入反应瓶中,开启搅拌,在室温下氮气置换三次。加热至90℃搅拌过夜后,将反应液与1000mL纯化水混合,抽滤,用二氯甲烷800mL溶解滤饼,有机相用1N盐酸溶液和1000mL纯化水洗涤,减压浓缩至干。加入50mL甲醇,在0℃下打浆1小时后抽滤,得到5-苯氧基异苯并呋喃-1(3H)-酮(22.5g,63%)。
第二步:2-(氯甲基)-4-苯氧基苯甲酸甲酯将1.0g 5-苯氧基异苯并呋喃-1(3H)-酮、10mL二甲苯、300mg苄基三乙基氯化铵和200mg三氟化硼乙醚加入反应瓶中,加热至100℃滴加5.0g二氯亚砜。滴完后继续加热至135℃反应5小时,薄层色谱监测反应完全。减压浓缩滤液,用甲醇(3mL)在55℃下搅拌反应1小时,浓缩至干,再用乙酸乙酯溶解,依次用水和氯化钠溶液洗涤。浓缩后得到2-(氯甲基)-4-苯氧基苯甲酸甲酯(1.0g,83.0%),直接用于下一步反应。
第三步:4-羟基-7-苯氧基异喹啉-3-甲酸甲酯将0.68g 2-(氯甲基)-4-苯氧基苯甲酸甲酯、0.7g N-对甲苯磺酰基甘氨酸甲酯、0.68g碳酸钾和0.16g碘化钾加入反应瓶中,氮气置换后加热至50℃搅拌1小时。降至室温后加入2mL甲醇钠甲醇溶液,继续搅拌30分钟。薄层色谱监测反应完全。反应液用40mL纯化水稀释,在搅拌下滴加冰醋酸调节pH值为7。抽滤得滤饼,加入4mL丙酮溶解后再次抽滤,得到4-羟基-7-苯氧基异喹啉-3-甲酸甲酯(525mg,72.5%)。
用途4-羟基-7-苯氧基异喹啉-3-甲酸甲酯是N-[(4-羟基-1-甲基-7-苯氧基-3-异喹啉基)羰基]甘氨酸(H948180)的中间体,是一种低氧诱导因子(HIF)脯氨酰羟化酶抑制剂。用于增加血液和骨髓造血祖细胞中的白细胞水平。
