氟代苯乙酸是一种重要的中间体,在制药、电子材料等合成过程中有着不可替代的作用。例如,间氟苯乙酸可用于抗炎药物的制备;2,4,5-氟代苯乙酸是制备糖尿病治疗药物西他列汀的关键中间体;而2,3-二氟苯乙酸则可用于制备二氟萘酚类液晶中间体。
制备 间氟苯乙酸的制备首先向反应瓶中加入350克32% HCl和250克3-氟苯胺,加热搅拌至溶液澄清。降温至-5~5℃后,依次添加20g四甲基氯化铵和20g氯化铜。随后滴加含300g偏二氯乙烯的丙酮溶液,并缓慢滴入含230g亚硝酸异丙酯和200g丙酮的混合液。维持温度下反应2小时后,淬灭反应。利用二氯甲烷进行萃取并蒸馏,最终收集到1-(2,2,2-三氯乙基)-3-氟苯398g。
接着,在氮气保护条件下,向三口瓶中加入500g水和60g对甲苯磺酸,并加热至80~95℃。滴加300g中间体1-(2,2,2-三氯乙基)-3-氟苯。滴加完成后,保温反应8小时。反应结束后将体系降温并加入冰水中淬灭。过滤、水洗后干燥,并用二氯乙烷进行重结晶,最终得到纯品间氟苯乙酸151.2g,其HPLC纯度大于99wt%。
