在装有气体吸收装置和回流冷凝管的100mL三口瓶中,加入反式-4-甲氧基苯基丙烯酸(17.8g,0.10mol)和甲苯(55mL),滴加氯化亚砜(11.9g,0.10mol)。缓慢加热至70℃,恒温反应4小时。冷却后,减压浓缩除去过量的氯化亚砜,残余物冷冻析出浅黄色固体,用二氯甲烷重结晶得到白色晶体(17.3g),收率88.0%。
化合物(4)的合成:在250mL三口瓶内加入四氢呋喃60mL,4-氨基苯硼酸(12.1g,88.0mmol)和三乙胺(13.3g,132.0mmol)。降温至-5~0℃下滴加溶于四氢呋喃(70mL)的化合物(3)(17.3g,88.0mmol),滴毕,室温搅拌2小时。用浓盐酸酸化反应液至pH值2-3,析出大量白色固体,过滤、水洗至中性后干燥得到24.6g白色固体产品,收率94.1%。
6-羟基-2(1H)-喹啉酮化合物(1)的合成:在500mL三口瓶内加入上一步产物(24.6g,82.7mmol)和甲苯250mL。降温至0℃下分批加入B(C6F5)3(2.2g,4.2mmol)和无水三氯化铝(15.4g,115.8mmol)。缓慢升温至90℃继续搅拌2小时后,将反应液倒入800mL冰盐酸溶液中使pH值2-3,搅拌1小时分层。保留有机层,并向所得有机层中滴加醋酸(49.6g,0.827mol),0~10℃下滴加30% H2O2(9.4g,82.7mmol)溶液,滴毕,0~10℃保温1小时。室温继续搅拌1小时后,滴加亚硫酸氢钠溶液淬灭过氧化物,分出有机层,饱和食盐水洗一次。有机层减压浓缩得到粗品固体经甲醇重结晶得到10.8g白色固体产品6-羟基-2(1H)-喹啉酮,收率80.7%,熔点300℃,纯度99.2%。
