Synthesis and anti-nociceptive and anti-inflammatory effects of gaultherin and its analogs

The synthesis of gaultherin (1) and its analogs was carried out to provide 11 glycosides under phase-transfer catalytic conditions. The activities of all synthesized compounds were evaluated by nitric oxide production inhibitory assay in vitro. Methyl 2-O-(4-O-beta-D-galactopyranosyl)-beta-D-glucopyranosylbenzoate (5f) showed significantly antinociceptive and anti-inflammatory effects by the evaluation in vivo. Structure-activity relationships within these compounds were discussed.

The present disclosure provides methods for obtaining gaultherin, a natural salicylate derivative, from plant tissue. The methods involve preventing the hydrolysis of the gaultherin in the plant tissue by disrupting the plant tissue under solvent conditions lacking a drying agent. The disclosure further provides various forms of plant-derived gaultherin, including alcoholic extracts, aqueous solutions and dried preparations, all suitable for use as a natural aspirin substitute. Moreover, the disclosure provides methods of using gaultherin-containing compositions to increase serum salicylic acid, to treat cancer cells, to treat cardiovascular (e.g., cardiopulmonary) conditions, to treat anti-inflammatory responses, and to inhibit cyclooxygenase.
A topical composition comprising a lipoic acid, a carnitine, and a carnosine in a suitable vehicle for topical application and a method for treating skin is provided. The present compositions are useful in improving the appearance of aged skin characterized by wrinkles and loss of elasticity. Preferred components include R-lipoic acid or R-dihydrolipoic acid, acetyl-1-carnitine, and 1-carnosine.
