ExoSensor 517: A Dual-Analyte Fluorescent Chemosensor for Visualizing Neurotransmitter Exocytosis

A dual-analyte fluorescent chemosensor (ExoSensor 517) for the direct visualization of neurotransmitters released upon exocytosis is presented. The sensor exploits the high concentration of neurotransmitters (e.g., glutamate, norepinephrine, and dopamine) and the pH gradient between the vesicle and synaptic cleft. The cooperative recognition elements require both binding and a change in environmental pH to afford a fluorescence response which makes ExoSensor 517 one of the first integrated molecular logic gates to be used for biological applications.
Thienochromene derivatives inhibit pSTAT1 and pSTAT5 signaling induced by cytokines

Furanocoumarins, particularly furo[3,2-c]coumarins, are found in many natural products. However, coumarins annulated to a thiophene ring have received scarce attention to date in the literature. Therefore, we synthesized 4-oxo-4H-thieno [3,2-c] chromene derivatives and tested in vitro their anti-inflammatory activity. Anti-inflammatory potential of the synthesized compounds (1, 2, 6-8, 9a-e and 10a-c) has been evaluated by measuring various pSTAT (signal transducer and activator of transcription) inhibition within the JAK (Janus-activated family kinase)/STAT signaling pathway. Ethyl 7-hydroxy-4-oxo-4H-thieno[3,2-cichromene-2-carboxylate (7) showed best inhibition properties on pSTAT5 in GM-CSF (Granulocyte-macrophage colony-stimulating factor)-triggered PBMC assay, with IC50 value of 5.0 mu M.
Pd-Catalyzed cross-coupling synthesis of 4-aryl-3-formylcoumarins

The threefold cross-coupling of triarylbismuthreagents with 4-chloro-3-formylcoumarins furnished the corresponding 4-aryl-3-formylcoumarins in a chemoselective manner with high yields under Pd-catalyzed conditions. This method was successfully applied to electronically different triarylbismuthreagents and 4-chloro-3-formylcoumarins preserving the 3-formyl group in the coumarin scaffold.
