制备6-溴茚酮可通过以下步骤制备:将三氟甲磺酸(0.35 mL,4 mmol)加入到酰胺底物6(1 mmol)的无水CHCl₃或CH₂Cl₂(2 mL)溶液中。搅拌混合物4小时后,将其倒入冰水中。将产物用CHCl₃萃取,并用水、盐水洗涤有机溶液,然后使用无水硫酸钠干燥。粗产物经过柱色谱(己烷:乙酸乙酯)纯化,回收硝基取代的苯胺。通过加入10M NaOH使含水萃取物呈碱性,并用CHCl₃萃取溶液。将有机溶液用水和盐水(2次)洗涤,再经无水硫酸钠干燥后得到6-溴茚酮,收率为88%。
