HIV-1 整合酶抑制剂 8 是一种 HIV-1 整合酶(HIV-1 integrase)的抑制剂,化合物 8。
体外研究在 Mn²⁺ 存在下作为阳离子辅因子时,HIV-1 整合酶抑制剂 8 可以有效抑制 3′-加工 (TC) 和 strand-transfer (ST) 活性。其 IC₅₀ 值分别为 275 µM 和 200 µM。它还抑制 strand-transfer (ST) 活性,IC₅₀ 值为 200 µM。
MCV 超螺旋酶的 DNA 松弛活性通过凝胶电泳监测;DNA 切割和连接活性则使用微量滴定板法进行检测。HIV-1 整合酶抑制剂 8 抑制 MCV 超螺旋酶以及 DNA 连接的 IC₅₀ 值分别为 500 µM 和 200 µM,这表明化合物 8 在这两种实验中对超螺旋酶都是无活性的。此外,HIV-1 整合酶抑制剂 8 诱导细胞毒性,在 HeLa 细胞中的 LD₅₀(导致信号降低 50% 的毒物剂量)为 20 µM。
