  A general method based on difference circular dichroic (DIF CD) spectroscopy for assigning the absolute configuration of 1,2,4-triol is presented. Four possible stereoisomers of 6-heptene-1,2,4-triol were prepared and served as models to develop the procedure. The sign of the DIF CD Cotton effect is correlated to the absolute configuration of the C2 position.

This convergent synthesis utilizes Vilsmeier-Haack formylation, Jacobsen hydrolytic kinetic resolution, esterification under Yamaguchi conditions and ring-closing metathesis as key steps.

  The absolute stereochemistry of the C-4 ∼ C-17 fragment of theonezolides was establsihed as 8S, 10R, 14S, 16S on the basis of synthesis of the two diastereomers (4a and 4b) suggested from model studies and comparison of their spectral and optical data with those of natural specimen (4) obtained from ozonolysis of 1 ∼ 3.
