美登醇是美登素类化合物(Maytansinoids)的共同母体,后者为酯类衍生物,普遍显示抗癌作用。美登醇已由日本学者通过发酵途径大量获得并转变成美登素类化合物。这类药物及其衍生物是微管组装极强的抑制剂,在亚纳摩尔水平即可表现出抗有丝分裂效应,且其ED50(有效剂量)在10-5~10-4μg/mL之间,是一种理想的抗癌药物。其中已上市的ADC药物中,罗氏的Kadcyla(2013年上市)的生物活性药物部分即采用了美登素类药物DM1。
美登醇制备以安斯菌素P3为原料制备美登醇。向反应瓶中加入100g AP3,并在-10℃至10℃氮气保护下,滴加浓度为1mol/L的LiAl(t-Bu)₃H四氢呋喃溶液。待LiAl(t-Bu)₃H四氢呋喃溶液滴加完毕后,在同样温度条件下搅拌2到5小时。反应液冷却至0℃至4℃,然后分批加入150ml水,并继续在-10℃至10℃下搅拌30分钟。随后,向其中加入200ml含有质量分数为1%甲酸的乙酸乙酯溶液,充分反应后过滤除去白色沉淀物,将滤液浓缩得白色泡沫固体。最后,在甲醇/二氯甲烷1%到4%梯度下进行柱层析,得到66g美登醇,产率为87%。
美登醇用途美登醇主要用于中药对照品、科研等领域。
生物活性Maytansinol可抑制微管组装并诱导微管解聚。与辐射联用时,可用于果蝇和人类癌细胞的研究。
