Regiospecific complexation of the dimethoxy arene ring of (–)-canadine to chromiumtricarbonyl, protection of the C-11 position, followed by stereoselectivesubstitution at C-8 effects, after deprotection and decomplexation, a stereoselectivesynthesis of (–)-(8R)-methylcanadine; a comparison is made with a racemic sample synthesised by an alternative route.

Each diastereoisomer of the C11-trimethylsilylcanadine complex may be treated with base and methyl iodide to give, after desilylation and decomplexatlon, the enantiomerically pure (-)-(8R) and (-)-(8S) methyl canadines; racemic samples of C8-methylcanadines are prepared an independent route.
