5-氨基吲唑可作为药物分子和有机合成中间体,并常作为基本骨架参与构建生物活性分子,例如HIV蛋白酶抑制剂1-(3-氨基吲唑-5-基)-3-苯甲基-环脲的合成。通过重氮化将苯环上的氨基基团转化为其他活性官能团,如卤素原子或叠氮基团等。
制备将5-硝基吲唑加入高压反应釜中,并用四氢呋喃溶解。随后,添加10% 碳钯;在25°C、5 kg压力的H₂大气下搅拌过夜。之后,使用N₂排出反应体系中的氢气并过滤混合物,再在真空条件下除去四氢呋喃溶剂以获得5-氨基吲唑(产物为棕色固体,产率为420 g,100%)。
