    An extensive study has been made on the antitumor activity of several iridoid glycosides and their aglycones in mice bearing the experimental tumor leukemia P388. Most of the aglycones were found to be active. Among them, scadoside methyl ester aglycone (SMEH) showed the most potent activity, and it was furhter tested against several kinds of experimental tumor. None of the glucosides showed any activity. Our observations suggest that the hemiacetal structure of iridoid aglycones plays a significant role in the antitumor activity, as in the case of the antimicrobial activity reported previously.

    compounds with chemical value, significantly simplifying the reaction work-up. From Aucuba japonica Thunb., an abundant natural terpenoid, aucubin, was isolated. Its reduced derivatives and corresponding unstable aglucons were purified by CPC. The latter chiral synthons were obtained under protecting group free conditions using eco-friendly, non-chlorinated solvents and without silica gel.

    A single crystal of iridoid related compound isoeucommiol was obtained through NaBH4 reduction of aucubigenin which was formed by enzymatic hydrolysis of aucubin. Its structure and stereochemistry were determined by X-ray diffraction. The crystal adopts an envelope conformation, presenting a triclinic system, space group P1 with Z = 1, unit cell dimensions a = 5.4158(13) Å, b = 6.5251(15) Å, and c = 7

  • X-ray crystal structure of iridoid glucoside aucubin and its aglycone

    X-ray diffraction analyses of iridoid glycoside aucubin (1) and its aglycone aucubigenin (2) are reported. it was found that crystals of 1 are orthorhombic, with P2(1)2(1)2(1) space group, both cyclopentane ring and pyran ring adopt envelope conformations, and the Glc moiety is in the C-4(1), conformation. Crystals of 2 are monoclinic, with space group P2(1), the cyclopentane and pyran rings also adopt the envelope conformation. The absolute configurations of 1 and 2 were also determined. Intensive O-H center dot center dot center dot O hydrogen bonds in both crystal lattices were observed. (C) 2009 Elsevier Ltd. All rights reserved.
  • Determination of Antibacterial Activity of Aucubigenin and Aucubin

    Because of the previously conflicted reports, the aucubigenin and aucubin have been reinvestigated for their antibacterial activities by a sensitive standardized micro-well dilution with automated spectrophotometric method, compared with paper disc diffusion method. The micro-well dilution assay was more accurate than paper disc diffusion method. The results showed that aucubin exhibited no antibacterial activity, while aucubigenin showed remarkable antibacterial activity against all of the tested bacteria. Aucubigenin was particularly more effective against Staphylococcus aureus with MIC value of 0.0313 mg/mL and MBC value of 0.0625 mg/mL, respectively.
