  Synthesis and in-vitro evaluation of 2-amino-4-arylthiazole as inhibitor of 3D polymerase against foot-and-mouth disease (FMD)

  Foot-and-mouth disease (FMD) is a highly contagious vesicular disease of livestock caused by a highly variable RNA virus, foot-and-mouth disease virus (FMDV). One of the targets to suppress expansion of and to control FMD is 3D polymerase (FMDV 3Dpol). In this study, 2-amino-4-arylthiazole derivatives were synthesized and evaluated for their inhibitory activity against FMDV 3Dpol. Among them, compound 20i exhibited the most potent functional inhibition (IC50 = 039 mu M) of FMDV 3D polymerase and compound 24a (EC50 = 13.09 mu M) showed more potent antiviral activity than ribavirin (EC50 = 1367 mu M) and T1105 (EC50 = 347 mu M) with IBRS-2 cells infected by the FMDV O/SKR/2010 strain.   In a homogeneous solution, the photo-Fries rearrangement of 2,5-disubstituted phenyl acetates gives ortho-hydroxyacetophenones as major photoproducts. The ortholpara ratio can be increased by using a highly viscous medium or restricted spaces such as those in zeolites. On the other hand, micellar solutions have little effect.

  This paper describes some progress made in helping a young aphasic man (MD) to overcome what had appeared to be intractable written language expression difficulties, which continued to be significant for him. Traditional therapy methods based on cognitive neuropsychological assessment had failed to help MD to generalize improvement in his spelling particularly, but not exclusively, of longer and irregular words. Both a splint which allowed him to use his dominant hand for writing directly onto a computer screen and a simple word-processing programme with synthesized auditory feedback and lexical and grammatical prediction (Write:OutLoud(R) and Co:Writer(R)) enabled him to produce more normal written output (increased quantity and more normal quality). The discussion also focuses on how effectiveness of language therapy can be measured in this man with chronic aphasia.
